The route
It's about 220 km on NH37, 4 to 5 hours depending on traffic. Compared to Shillong or Cherrapunji this one's easy, flat road, not much turns, nothing that really slows a car down.
You can do it as a day trip if you're okay with a long one. Leave by 5 or 6, you're in Kaziranga around 10, get a safari done, back in Guwahati by evening. People do it this way all the time. But it's a full day sitting in a car either direction, so if you'd rather not rush it, stay a night near the Kaziranga national park instead. You get an evening there, not just a few rushed daylight hours.
The national park mostly open in the month of October through April, shut the rest of the time for monsoon flooding. October and the March-April stretch usually have the best weather and fewer crowds than peak winter. Safaris are subjected to weather conditions so it might be close if there is extreme rainfall.

Safaris and where to stay
In Kaziranga, safaris can be done from different ranges. Kohora, Bagori, or Agoratoli being the most famous, each a bit different. Early slot, 6 AM, is generally the one to book if sightings matter to you, animals are more active before the jeeps start moving through.
